TRIUMPH Program

TRIUMPH stands for “Triple Receptor Agonist Intervention Utilizing Metabolic Pathways in Health.” It is Eli Lilly’s Phase 3 clinical development program for retatrutide in obesity and weight management. The program is anchored by four pivotal trials, each addressing a distinct obesity subpopulation, plus a separate cardiovascular outcomes trial:

• TRIUMPH-1: obesity or overweight with at least one weight-related comorbidity, no type 2 diabetes (n=2,339; reported May 21, 2026; -28.3% mean weight loss at 12 mg/80 weeks)
• TRIUMPH-2: obesity with type 2 diabetes, with a nested obstructive sleep apnea basket (readout expected Q2-Q3 2026)
• TRIUMPH-3: Class II/III obesity (BMI ≥35) with established cardiovascular disease (readout expected Q3-Q4 2026)
• TRIUMPH-4: obesity with knee osteoarthritis (n=445; reported December 2025; -28.7% mean weight loss at 12 mg/68 weeks)
• TRIUMPH-Outcomes: ~10,000-patient cardiovascular outcomes trial (CVOT) with MACE as primary endpoint; multi-year duration, post-approval

The TRIUMPH name and trial population breakdown distinguish it from the parallel TRANSCEND program (type 2 diabetes glycemic control as primary indication) and the SYNERGY program (metabolic dysfunction-associated steatohepatitis, or MASH).
